چکیده

Although the auger-like 'swimming' motility of the African trypanosome was described upon its discovery over one hundred years ago, the precise biomechan-ical and biophysical properties of trypanosome flagellar motion has not been elucidated. In this study, we describe five different modes of flagellar beat/wave patterns in African trypanosomes by microscopically examining the flagellar movements of chemically tethered cells. The dynamic nature of the different beat/wave patterns suggests that flagellar motion in Trypanosoma brucei is a complex mixture of oscillating waves, rigid bends, helical twists and non-linear waves. Interestingly , we have observed soliton-like depression waves along the flagellar membrane , suggesting a nonlinear mechanism for the dynamics of this system. The physical model is inspired by the 2-dimensional elastic dynamics of a beam, and by taking into account uniform distribution of molecular motors torque and non-linear terms in the curvature.

Direct Measurements of the Stiffness of Echinoderm Sperm Flagella

1. The stiffness (flexural rigidity) of some echinoderm sperm flagella was measured, using a flexible glass microneedle. 2. Values of 0-3-1 -5 x io a i N m were obtained for the stiffness of live flagella which were immobilized with COg-saturated sea water. 3. The immobilized live flagellum was uniform in stiffness along its entire length, except in a particular plane of imposed bending in whic...

Multicellularity and the functional interdependence of motility and molecular transport.

Benefits, costs, and requirements accompany the transition from motile totipotent unicellular organisms to multicellular organisms having cells specialized into reproductive (germ) and vegetative (sterile soma) functions such as motility. In flagellated colonial organisms such as the volvocalean green algae, organized beating by the somatic cells' flagella yields propulsion important in photota...

Visualization of flagellar interactions on bacterial carpets.

Methods for the in-depth study of the physics of microscale actuation of microfluidics environments by flagellated bacteria 'teamsters' have been developed. These methods, which include single and multi-colour fluorescent labelling and electron microscopy allow for the analysis of the effect that individual flagellar filaments have on bacterially driven microstructures, and allow for the invest...

Functional expression of adhesive peptides as fusions to Escherichia coli flagellin.

An expression system for studying epitopes of adhesion proteins based on fusion of gene fragments into fliC(H7) of Escherichia coli is described. We constructed the system by an in-frame insertion of DNA fragments encoding one, two or three of the fibronectin-binding D repeats present in the fibronectin-binding protein A (FnBPA) of Staphylococcus aureus, into the fliC(H7) gene region encoding t...
